Hello,
Is 375 load rating (four casters) sufficient to support the chassis until I put the wheels and tires on?

Those are some pretty lightweight wheels though.

You need to clarify what build stage the chassis is/would be at. Is it:
- bare (yes)
- roller (likely)
- or mostly finished with the drivetrain, etc. installed (no)?

I have 3 body bucks and a chassis dolly that all get used constantly. After having been through a variety of caster setups I only have one word for you: PNEUMATIC TIRES! OK, make that two words. They roll easily, tolerate uneven surfaces and won't be stopped dead when a wheel encounters a 1/4" nut on the floor like will happen with the smaller diameter hard wheels. I like at least 8" but don't rule out 10" or 12". After having set the bucks and dolly up in various configurations with 2 and 4 swivels I prefer 2 fixed wheels and 2 swivel---although it sounds counterintuitive 4 swivels can be more difficult to maneuver because when you swing one end the other end usually tries to go the opposite direction. Pretty good selection available at Harbor Freight, Amazon, etc. for cheap money. Something like this:
