I have a new Tremec TKO500 transmission with stock shifter. IYO is it worth while to replace the stock shifter with an after market one? Will it shift smoother?

Tremec in general is quite hard and distinct to shift, I dont think it help to use another shifter but It will be some better over time...

I did buy an after market shifter. I decided to go with a Steeda shifter. The Steeda shifter seems to be much better. It shifts much crisper between gears especially the shift between 2nd & 3rd gears.

i bought the steeda one also, more positive feel, slightly shorter throw,

Have a TKO-600 that shifts very well. Don't think its the shift handle but FWIW I do have the aftermarket 427 version with the fake Tee reverse lockout pull from **********.

I also have the aftermarket 427 version shifter. I installed that one first on the Steeda Triax shifter, but I liked the shift lever that came with the Steeda Shifter. It has a nice offset and a shorter throw. Although the 427 shift lever looks better. I did install an aftermarket leather shift boot that looks way better than the FF supplied one.
